KSM-66® Ashwagandha · 300 mg per serving
In a 60-day randomized, double-blind, placebo-controlled trial of 64 adults with a history of chronic stress, participants took 300 mg of a high-concentration full-spectrum ashwagandha root extract twice daily. The treated group showed a 27.9% reduction in serum cortisol from baseline, against 7.9% in the placebo group, alongside significant improvements across the Perceived Stress Scale, GHQ-28 and DASS. Adverse effects were mild and comparable between groups.

L-Theanine · 200 mg per serving
A four-week randomized, double-blind, placebo-controlled crossover trial gave 200 mg of L-theanine daily to 30 healthy adults, 21 of them women, with a mean age of 48. Scores on the Self-rating Depression Scale, trait anxiety and the Pittsburgh Sleep Quality Index all decreased on L-theanine and not on placebo. A separate crossover trial found the salivary cortisol response to an acute cognitive stressor was reduced three hours after a 200 mg dose.

Rhodiola Rosea · 150 mg per serving
A phase III randomized, double-blind, placebo-controlled trial enrolled 60 adults aged 20 to 55 who met diagnostic criteria for stress-related fatigue. Over 28 days, the rhodiola group improved significantly against placebo on Pines’ burnout scale and on several attention measures, and the cortisol response to awakening differed significantly between groups from pre- to post-treatment. The authors concluded the extract exerts an anti-fatigue effect that increases the ability to concentrate.

Phosphatidylserine · 50 mg per serving
Phosphatidylserine has been studied for its effect on the hypothalamic-pituitary-adrenal axis under load. In an early placebo-controlled study, chronic administration blunted the ACTH and cortisol responses to physical stress. A later randomized, placebo-controlled study of a soy-based phosphatidylserine and phosphatidic acid complex reported normalized HPA-axis stress reactivity in chronically stressed subjects. Effects in this literature are dose-dependent, and the doses studied vary widely.

Magnesium Bisglycinate · with B6 and D3
A 2020 review in Nutrients describes the relationship between magnesium and stress as a self-reinforcing loop: stress increases magnesium excretion, and lower magnesium status in turn heightens stress reactivity. The authors note that magnesium supplementation, particularly alongside vitamin B6, has been examined as a way to interrupt that cycle. We use the bisglycinate form, chelated to glycine, which is better tolerated on a light stomach than magnesium oxide.
